李筝,广州国家实验室基础研究部研究员,广州医科大学特聘教授,国家重大人才项目(青年)入选者。2016年于中国科学院大学(上海生化细胞所)获得博士学位。2016-2020年,在耶鲁大学完成博士后训练,主要从事DNA修饰介导的表观遗传调控机制与功能方面研究。2020-2021年在生物岛实验室担任研究员,现任广州国家实验室研究员,主要聚焦DNA修饰等表观遗传因子在呼吸系统疾病发生发展中的作用和分子机制研究。具体包括:呼吸系统疾病的表观遗传调控机制;肺细胞分化过程中的表观遗传调控;基因组上DNA修饰建立与调控的分子机制等。代表性研究成果以第一作者/共同第一作者身份发表在Nature、Nucleic Acids Research等国际期刊上,亦作为合作作者在Cell、Science、Cell Stem Cell及Cell Research等杂志发表多篇论文。
报告摘要:
Totipotent cells possess the unique ability to generate all embryonic and extraembryonic lineages. Although various protocols have been established to induce the transition from pluripotency to totipotency, the molecular mechanisms underlying this process remain incompletely understood. Here, we identify the RNA-binding protein Embryonic Lethal Vision-Like Protein l (Elavll) as a critical regulator of totipotent cell induction. Loss of Elavll impairs the acquisition of totipotency without disrupting pluripotency maintenance. Mechanistically, Elavll acts beyond its canonical role in mRNA stability to directly regulate the transcription of totipotency-associated genes, including the key transcription factor Dux. We show that Elavll facilitates the activation of these genes through an RNA-dependent chromatin-binding mechanism. This interaction enhances local chromatin accessibility and drives transcriptional upregulation of totipotency-related programs. Our findings uncover a previously unrecognized RNA-mediated pathway by which Elavll interfaces with chromatin to orchestrate cell fate transitions, providing new insights into the molecular control of totipotency.
